Coordination Mode of Hesperidin with Cu(II) and the Antioxidation Mechanism of Hesperidin
The coordination mode of hesperidin with Cu(II) and the antioxidation mechanism of hesperidin were investigated by combined use of the Complete Neglect Differential Overlap CNDO calculation mole ratio method conductivity method, method of continual variations of equimolar solutions, and IR spectrum analysis. The results show that hesperidin can not scavenge hydroxyl radicals (HO·) effectively because it contains only two phenol hydroxyls. However it can form a complex of 21 with Cu(II) through the carbonyl group and the ortho-hydroxyl group. The relative stable constant lgK of the complex is 8.05. In addition, H is given off during the coordination. So in the generating system of free radicals arising when Cu(II) catalyzes the decomposition of hydroperoxides, hesperidin functions as an antioxidant mainly by chelating Cu(II) to prevent hydroperoxides from being decomposed into free radicals, whereas its hydroxyl radicals scavenging effect is much less important.
